    “…Objective To investigate the role of caveolae on vascular smooth muscle cells(VSMCs)in vasoconstriction of renal artery(RA)and the possible mechanisms.Methods The RA was pretreated with methyl-beta-cyclodextrin(MCD)and cholesterol(CHO)to reduce and enhance the formation of caveolae,respectively.Isometric force recording system was used to detect the vascular function.Real-time polymerase chain reaction(RT-PCR) and Western blotting were conducted to examine the expression of mRNA and protein of caveolin-1 and Angiotensin type 1 receptor(AT1R).Immumoprecipitation with Western blotting was carried out to test the binding of caveolin-1 and AT1 R.Results Incubation with MCD reduced the caveolae number in VSMCs significantly(P<0.05),which could be reversed by CHO treatment.Neither the MCD nor CHO influenced the gene or protein expression of caveolin-1.The vasoconstriction of RA to KCl or PE did not change after MCD or CHO incubation.However,MCD could inhibit the vasoconstriction of the artery to Ang Ⅱ with lower Emax(P<0.05)and higher EC50(P<0.05),which again could be reversed by CHO pretreatment The protein expression of AT1 R was not affected by MCD or CHO incubation.In the case of normal RA,Ang Ⅱ would promote the binding of AT1 R to caveolin-1(P<0.05),while the response was blocked substantially in MCD-incubated RA,which was restored if the RA was treated by MCD and CHO simultaneously.Conclusions The caveolae regulate the vasoconstriction of RA to Ang Ⅱ by controlling the binding of caveolin-1 with AT1 R.…”
