    “…Objective To investigate the effect of simvastatin on the DAG/PKC and MAPK signaling pathways in type 2 diabetic nephropathy patients.Methods 16 patients with diabetic nephropathy were treated from June 2010 to December 2014 in Huangshi Central Hospital.These patients given simvastatin.After treatment for 3 months,15 mL blood was collected from each patient.Automatic biochemical analyzer was used to determine renal function parameters,and a kit was used to detected serum lipids,IL-1α,IL-6,IL-1β and TNF-a.The expression of DAG/PKC and MAPK signaling pathway-related enzyme proteins was compared before and after treatment.Results After treatment,blood urea nitrogen,serum creatinine and serum uric acid were reduced significantly in patients after treatment as compared with in patients before treatment(P<0.05).The endogenous creinine clearance rate in patients after treatment was significantly higher than in patients before treatment(P<0.05).CHOL,TG and LDL-C levels in patients after treatment was significantly lower than in patients before treatment(P<0.05).The HDL-C level in patients after treatment was significantly higher than in patients before treatment The levels of inflammatory cytokines(IL-1α,IL-1β,IL-6and TNF-a) were significantly lower in patients after treatment than in patients before treatment(P<0.05).The expression of PKC-η and PKC-ζ protein and mRNA was significantly lower in patients after treatment than in patients before treatment The expression levels of MAPK,MKK,and MAKK protein and mRNA in patients after treatment was significantly lower than in patients before treatment(P<0.05).Conclusions Simvastatin can improve renal function in type 2 diabetic nephropathy patients by regulating DAG-PKC and MAPK signaling pathway.…”
