    “…Objective To explore the expression level of CX3CL1 and CX3CR1 in renal tissue of chronic kidney disease(CKD) patients and unilateral ureter obstruction(UUO) mice and their association with renal fibrosis.Methods(1) The expression of CX3CL1 and CX3CR1 in renal biopsy of 30 initially diagnosed CKD patients and 10 healthy controls was detected by immunohistochemistry.(2)Western blotting and immunohistochemistry were respectively performed to detect the expression level of CX3CL1 and CX3CR1 in renal tissues of wild-type(WT) mice and UUO model at 7th day and 14 th day.(3) The levels of renal fibrosis and macrophage infiltration in the CX3CR1<sup>-/-</sup> knockout and widetype UUO model mice were evaluated by immunofluorescence.Results The expression of CX3CL1 C and CX3CR1 in renal tissues of initially diagnosed CKD patients was significantly higher than that in healthy controls(P<0.05).Besides,the levels of CX3CL1 and CX3CR1 in UUO models were higher than in wild-type mice(P<0.05).Compared to the 7-day UUO models,the 14-day UUO models had significantly higher level of CX3CL1 and CX3CR1 in renal tissues(P<0.05).Compared to wide-type UUO models,the levels of orSMA and macrophage infiltration in CX3CR1<sup>-/-</sup> knockout models were significantly decreased(P<0.05).Conclusions Our study indicated that the expression levels of CX3CL1 and CX3CR1 in initially diagnosed CKD patients and UUO mice were obviously increased in renal tissues.The overexpression of CX3CL1 and CX3CR1 in renal tissues was probably closely related with renal fibrosis.…”
