    “…Objective To study the effect of Huangkui capsule inhibiting oxidative stress and fibrosis index in patients with diabetic kidney disease(DKD)with stageⅢ-Ⅳ,so as to explore the mechanism of Huangkui capsule in the treatment of DKD.Methods 100 patients with DKD with stageⅢ-Ⅳfrom June 2014 to October 2015 were randomly divided into two groups,50 in each group.The patients in the control group were given conventional treatment,and those in the treatment group additionally received Huangkui capsule orally,5 capsules each time,3 times every day for 6 months.The kidney function,and urine and serum levels of oxidative stress and fibrosis indicators were detected.Results The SCr,BUN,andβ2-MG levels in two groups after treatment decreased,CCr increased,and there was statistically significant significance between two groups(P <0.05).The UAER and 24-h urine proteinin two groups after treatment were significantly lower than those before treatment,and those in the treatment group were significantly lower than those in the control group(P<0.05 for all).The MDA and MPO concentrations intwo groups after treatment were significantly lower than before treatment,SOD activity was significantly higher than before treatment,and there was statistically significant difference between two groups(P<0.05).There was no statistically significant difference in 8-OHdG in two groups before and after treatment(P>0.05).The serum IV-C,PC Ⅲ,LN and HA levels after treatment in two groups were significantly lower than before treatment,and those in study group were significantly lower than those in the control group(P<0.05 for all).Conclusions Huangkui capsule can antagonize the oxidative stress injury in kidney tissues probably by influencing the release of fibrosis factor and improving the renal fibrosis.…”

    “…Objective To explore the role and mechanism of podocytes in rat model with uric acid-induce renal damage.Methods By using a random number table, a total of 24 male Sprague-Dawley rats were randomly divided into three groups of control(NC),model(HUA)and febuxostat(FX)(n=8 each).After 8 weeks, the rats were weighted and sacrificed.The levels of uric acid(UA),blood urea nitrogen(BUN),serum creatinine(Scr)and 24 h urinary protein(24 h UP)were recorded.Renal pathological changes were observed under transmission electron microscope(TEM)after hematoxylin-eosin(HE)staining.The renal expression levels of nephrin and desmin were detected by immunohistochemical staining and Western blot.Results At Week 8,the levels of UA,Scr, BUN,24 h UP and the expression level of desmin increased markedly in HUA group than those in NC group(P<0.01);The expression of nephrin decreased obviously in HUA group.Compared with HUA group, the levels of UA,Scr, BUN,24 h UP and the expression level of desmin decreased obviously in FX group.Massive urate crystals deposited in renal tubules and foot process effacement in HUA group.However, it was lighter in FX group.Conclusion Hyperuricemia causes renal podocyte damage and impairs renal function.Febuxostat treatment can effectively lesson the degree of podocyte damage in hyperuricemia rats.A positive correlation exists between serum uric acid level and desmin expression in renal tissue(P<0.01);a negative correlation between serum level of uric acid and nephrin expression in renal tissue(P<0.01).A linear correlation exists between the level of uric acid and the degree of podocyte injury.Podocyte injury may be one of the causes of uric acid nephropathy.…”

    “…Objective To explore the effects of anti-platelet membrane glycoprotein Ib alpha(GPIbα)antibody on the expression of peripheral GPIbα,alpha particle membrane glycoprotein(CD62 P)and lysosomal membrane protein(CD63)in maintenance hemodialysis(MHD)patients.Methods Thirty-seven patients with chronic renal failure undergoing MHD from March 2016 to March 2018 in the Department of Nephrology in Eighth Affiliated Hospital of Sun Yat-sen University and 31 healthy people in the Physical Examination Center were enrolled in this study.Using flow cytomery,the expression of platelet member GPIbα,CD62 P and CD63 was detected before and after addition of anti-GPIbαantibody in MHD patients.Results The expression of platelet membrane GPIbαin no antibody treatment group(29.9±4.9)% was significantly lower than in control group(63.8±9.0)%and antibody treatment group(42.3±6.5)%(both P<0.01).The expression of CD62 P and CD63 in no antibody treatment group(50.9±7.5)% and(54.1±5.5)% was significantly higher than that in the control group(23.3±8.7% and 30.9±6.2%)and antibody treatment group(36.5±2.0)% and(47.2±2.2)%(P<0.01,P<0.05).Conclusions Anti-GPIbαantibody can regulate platelet function and activity by up-regulating the expression of platelet membrane GPIbα,and down-regulating the expression of CD62 P and CD63 in MHD patients,as a result to suppress adhesion and aggregation of platelets and thrombosis,and reduce the possibility of thromboembolism events in MHD patients.…”

    “…Objective To investigate the role of caveolae in higher vasoconstriction of interlobar renal artery(IRA) of spontaneously hypertensive rat(SHR) to angiotensin Ⅱ(Ang Ⅱ).Methods Isometric force recording system was used to detect the vascular function.Methyl-beta-cyclodextrin(MCD) was employed to reduce the caveolae abundance.Real-time polymerase chain reaction(RTPCR) and Western blotting were conducted to examine the expression of mRNA and protein of angiotensin type 1 receptor(AT1R).Immumoprecipitation followed by Western blotting was carried out to test the binding of caveolin-1 and AT1 R.Results As compared to Wistar Kyoto(WKY) rats,the constriction of IRA to Ang Ⅱ was increased by 32.7%in SHR(P<0.05).Meanwhile,the abundance of caveolae on vascular smooth muscle cells(VSMCs) was enhanced significantly(P<0.05).After reducing the caveolae by MCD,the intensity and sensitivity of vasoconstriction to Ang Ⅱ in IRA of SHR were partly and entirely reversed,respectively.Moreover,AT1 R protein expression was higher in IRA of SHR group than in WKY group in which the AT1 R protein expression was lowered in part by MCD incubation.In IRA of both WKY group and SHR group,Ang Ⅱ could promote the binding of AT1 R to caveolin-1(P<0.05),which was by a larger extent in SHR group than in WKY group(P<0.05).After treatment with MCD,the binding of the two proteins in IRA of SHR group was restored to the level of WKY rats.Conclusions Caveolae regulates the enhanced intensity and sensitivity of vasoconstriction to Ang Ⅱ in IRA of SHR by raising AT1 R protein content and binding of AT1 R to caveolin-1.…”
