Summary:The principle of ATP bioluminescence assay and the measurement method were introduced. The method was applied to the measurement of micro-biomass in West Lake. The concentration of microorganisms in West Lake was 10<sup>6</sup>-10<sup>7</sup> cells·mL<sup>-1</sup>. In general, the concentration near bank was higher than that in the center of lake, and the concentration in summer was higher than that in spring. Human activities, such as tourism and angling, great influenced the microbial distribution in the lake. Moreover, the concentration of microorganisms in the vicinity of fishpond of a restaurant was relatively high, however, it had no obvious influence on the main body of lake. The import of pure water source was beneficial to the purity of water.
