| Summary: | Objective To discuss the intervention effects of Eicosapentaenoic acid on human albumin(Alb)-induced human proximal renal tubular epithelial cells(HK-2) transdifferentiation.Methods In vitro human proximal renal tubular epithelial cells were randomly divided into the following six groups:blank control group;30 μmol/L EPA group;5 mg/mL Alb-induced group;Alb + 10 μmol/L EPA group,Alb + 30μmol/L EPA group,and Alb + 50 μmol/L EPA group.Immunofluorescence was used to detect the protein expression of E-cadherin and alpha(α)-SMA ELISA was applied to detect FN protein level RT-PCR was employed to detect TGF-β1 mRNA expressioa Results As compared with group A,there was no significant difference in the expression of TGF-β1,α-SMA,FN and E-cadherin between groups A and B.The expression levels of TGF-β1,α-SMA and FN in group C were significantly higher,and those of E-cadherin obviously lower than other groups.The expression of TGF-β1,α-SMA and FN was reduced gradually,and that of E-cadherin increased gradually in groups D,E and F as compared with the group C,and the intra-group comparison showed significant difference.In short,EPA could significantly inhibit the expression of E-cadherin and α-SMA in HK-2 stimulated by Alb,and reduce the FN levels in the supernatant fluid in a concentration-dependent manner.EPA also significantly reduced the expression of TGF-β1.Conclusions A certain concentration of human Alb can induced epithelial-mesenchymal transition of in vitro cultured HK-2 cells.EPA can inhibit the transdifferentiation of HK-2 cells stimulated by Alb to some extent in a dose-dependent manner. |
