description [Objectives] To investigate the expression and patient prognosis of VSIG4 in colorectal cancer (CRC) and explore the relationship between VSIG4 expression and immune cell infiltration in CRC. [Methods] This study compiled and analyzed VSIG4 mRNA expression data from various public databases. Additionally, GO, KEGG, and GSEA enrichment analysis were used to further explore the biological role and signaling pathways of VSIG4 in CRC. Subsequently, the CIBERSORT algorithm was used to analyzed relationship between VSIG4 and immune cell infiltration, and the impact of VSIG4 expression on the prognosis of CRC patients was further discussed. [Results] Utilizing a total of 3 524 samples, the meta-analysis substantiated the down-regulation of VSIG4 in CRC with a standard mean difference (SMD) of -0.60. Receiver operating characteristic (SROC) curve analysis suggested that VSIG4 effectively distinguishs CRC samples from non-cancer control samples (AUC=0.75). GO, KEGG, and GSEA enrichment analysis proposed that VSIG4 primarily regulates tumor immune responses by modulating the binding and activity of signaling receptors. CIBERSORT analysis revealed significant differences in immune cell infiltration between VSIG4 high/low expression groups in CRC samples. Drawing on a total of 970 CRC patient survival data, the meta-analysis suggested a significant association between VSIG4 expression and CRC patient prognosis. [Conclusion] This study affirmsthe down-regulation of VSIG4 in CRC and establishes its role in CRC development through the regulation of immune responses, consequently influencing the patient prognosis.
