Using a direct high frequency shoot regeneration system suitable for genetic transformation was established successfully in this study, which was the base for the following research of construction of a direct high frequency shoot regeneration system suitable for genetic transformation was established successfully by using pieces of fresh stem of Muscat of Alexandria (Vitis vinifera L. var. muscat) as explants, and transformed plants were obtained with a higher transformation efficiency. Stem pieces from in vitro-grow plants were cocultivated for 3 days with Agrobacterium tumefaciens strain GV3101 containing the vector pDC202 in which LEAFY gene from Arabrodopsis has been inserted. It was followed by incubating on regeneration medium (GS+ZT 2.0 mg·L<sup>-1</sup>) containing rif, and gen for 4 weeks, After observed according its growth status and identified by use of PCR and Southern blot, the transformed plants was obtained with roots growing at first and the buds at second, which was different from some other trarisgenic plants to some extent, and the regeneration percentage of transformed buds was 6.8%.
