Summary:To study insecticidal mechanism of terpinen-4-ol, the adults of Musca domestica were investigated with terpinen-4-ol by topical application. The activities of carboxyl esterases (CarE), phosphatase, glutathione S-transferase (GSTs), cytochrome P450 (P450), and polyphenol oxidase (PPO) of insects tested were determined in all poisoning stages, including exciting stage, convulsing stage, paralysis stage, and recover stage. The result shows that the activity of CarE in houseflies tested was induced by terpinen-4-ol. Acid phosphatase (ACP) was induced in exciting stage, convulsing stage, and recovering stage, but was inhibited in paralysis stage. The activity of alkaline phosphatase (AKP) was inhibited, the inhibition rates were 8.0%, 28.0%, 45.0%, and 33.0% in exciting stage, convulsing stage, paralysis stage, and recovering stage, respectively. GSTs in houseflies treated were induced, and the specific activities in all four poisoning stages were 1.59, 1.19, 1.26, and 2.76 times of the control. O-demethylase activity of cytochrome P450 was inhibited by terpinen-4-ol, and the inhibition rates in paralysis stage and recovering stage were 46.91% and 60.89%, respectively. PPO activity was strongly induced by terpinen-4-ol, both in v itro and in v ivo. In conclusion, the activities of metabolizing enzymes could have relation with toxicity of terpinen-4-ol against the adults of M. domestica, but the recover of the poisoning insects might be related to GSTs induced. As a new insecticide or synergist, terpinen-4-ol has a potential value in the field of insecticide resistance management.
