Summary:Background: Acute kidney injury (AKI) causes high mortality. Cell therapy has provided acceptable results, but factors such as immune rejection and teratoma formation have limited their use. The purpose of this research is to study the reparative effects of secretome secreted from kidney-derived stem cells on glycerol-induced kidney damage in rats. Materials and Methods: Secretomes were isolated from conditioned media of kidney mesenchymal stem cells. The animals were separated into 4 groups: Control, Secretome (received 30 μg/kg secretome, for 3 days), Glycerol (10 ml/kg of glycerol, intramuscularly) and Glycerol + Secretome (received glycerol and secretome). Blood urea (BUN), Creatinine (Cr) and Albumin (alb) levels were measured with detection kits. The histopathological results of the secretomes on kidney were checked with PAS staining and the amount of expression of miR-375 and miR-494 was assessed by quantitative real-time polymerase chain reaction (qRT-PCR). Results: The results of this research showed that intravenous of secretomes significantly decreased the levels of BUN (P < 0.01), Cr (P < 0.001), and alb (P < 0.001). Well as there was a considerable decreased in miR-375 (P < 0.001) and miR-494 (P < 0.01) in the receiving group of Glycerol + Secretome rats. The results showed secretomes improved kidney histopathology by inducing regulatory changes in the expression levels of microRNAs (miRNAs). Conclusion: The results of this research showed that secretomes extracted from kidney stem cells can play a role in reducing kidney damage by regulating miRNAs related to AKI.
