紫外-LiCl联合ARTP诱变选育高产花生四烯酸的 高山被孢霉突变株Breeding of Mortierella alpina mutants with high yield of arachidonic acid by UV-LiCl combined with ARTP mutagenesis
为得到遗传性状稳定、高产花生四烯酸(ARA)的高山被孢霉菌株,以实验室保藏的高山被孢霉FD-1为出发菌株,采用紫外-LiCl联合常压室温等离子体(ARTP)技术进行诱变处理,并进行TTC平板初筛和摇瓶发酵复筛。结果表明,经紫外-LiCl联合ARTP诱变处理,获得一株高产ARA的高山被孢霉突变株Z6-A23。菌株Z6-A23所产油脂中ARA含量高达44.57%,ARA产量达3.62 g/L,分别较出发菌株FD-1提高了75.40%和223.21%。经10次传代培养,突变株Z6-A23代谢合成ARA性能稳定,说明其具有良好的遗传稳定性。综上,突变株Z6-A23有望成为高产ARA的潜力菌株。In o...
