description | 目的探索霍乱弧菌新发菌株H1的六型分泌系统(T6SS)存在的转录调控机制。方法本研究以转录组测序为基础,分析H1株分别由于<italic>vipA</italic>敲除(H1Δ<italic>vipA</italic>),<italic>vipB</italic>敲除(H1Δ<italic>vipB</italic>)和<italic>clpV</italic>敲除(H1Δ<italic>clpV</italic>)时T6SS基因簇的转录水平,并对其差异基因进行了基因本体论(GO)富集和京都基因与基因组百科全书(KEGG)富集。结果发现H1Δ<italic>vipA</italic>有22个差异基因(FDR<0.01,|log<sub>2</sub>FC|≥1),其中有18个属于T6SS基因簇;并且除<italic>vipA</italic>外,其转录水平均呈上升趋势。结合GO富集和KEGG富集结果,发现<italic>vipA</italic>的缺失会引起与霍乱弧菌群体感应相关基因(<italic>hapR</italic>)的转录水平显著降低。结论我们得出结论<italic>vipA</italic>基因在霍乱弧菌H1菌中是一个具有多重功能的基因,一是编码T6SS鞘结构蛋白,二是参与T6SS调控;推测<italic>hapR</italic>可能参与H1株中<italic>vipA</italic>对T6SS基因簇的转录调控。 |
