Summary:Whitish muscle of Macrobrachium rosenbergii was a new serious disease occurred in recent years in the main region of giant freshwater prawns in China, and M. rosenbergii Nodavirus (MrNV) was confirmed as the pathogen of the diseases. The rabbit antiserum and monoclonal antibody 2B5 against MrNV, purified with ultra-speed centrifugation from diseased prawn postlarvae, were developed and used for the establishment of TAS-ELISA. The optimal condition of TAS-ELISA: rabbit antibody used for plate precoating was 1 μg·mL<sup>-1</sup>, work concentration of Mab 2B5 was 1∶50000. The sensitivity of TAS-ELISA to purified MrNV was 0.98 ng, without cross reaction to WSSV, TSV and other dead prawn postlarvae caused by bacterial infection. The positive detection rate as well as coincident rate were compared with different methods and showed that TAS-ELISA was better than indirect ELISA and RNA characterization with electrophoresis, when the diseased and suspicious prawn samples were analysed. Besides, the easy TAS-ELISA was developed with lower temperature and shorter incubation time, which made TAS-ELISA more practical in field detection.
