description To optimize the extraction process of polysaccharides from Ganoderma lucidum fruit body and preliminarily study its effect on asthmatic mice, using the extraction rate of polysaccharides from G. lucidum fruit body as response value, on the base of single-factor experiments, the polysaccharides extraction process was optimized by response surface methodology. Asthmatic mice model was induced by ovalbumin. Using dexamethasone (5 mg/kg) as the positive control, the effects of low-, medium- and high-dose polysaccharides from G. lucidum fruit body (50 mg/kg, 100 mg/kg, 200 mg/kg) on the pathomorphology of lung tissue, white blood cells and cell classification count, and the levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in bronchoalveolar lavage fluid were investigated. The results showed that the optimal extraction process of polysaccharides from G. lucidum fruit body was ultrasound power 400 W, time 40 min, and material to liquid ratio 1∶15 (g∶ml). Under these optimal conditions, the extraction rate of polysaccharides was (3.22&#x00B1;0.04)%. Compared with the model group, the pathological changes such as obvious thickening of blood vessels and bronchial walls in the lung tissue of asthmatic mice and massive infiltration of inflammatory cells were significantly improved in the high-dose G. lucidum polysaccharides group, and the numbers of white blood cells, lymphocytes, neutrophils, and eosinophils and the concentrations of IL-6 and TNF-α in bronchoalveolar lavage fluid were significantly reduced to (34.96&#x00B1;5.81)&#x00D7;10<sup>4</sup> CFU/ml, (25.61&#x00B1;3.59)&#x00D7;10<sup>4</sup> CFU/ml, (4.67&#x00B1;1.59)&#x00D7;10<sup>4</sup> CFU/ml, (3.06&#x00B1;1.131)&#x00D7;10<sup>4</sup> CFU/ml, (82.62&#x00B1;14.07) pg/ml and (208.16&#x00B1;19.60) ng/L, respectively. The results indicated that the polysaccharides from G. lucidum fruit body could improve the inflammatory response in asthmatic mice.
