Summary:Gene expression profile related to seed stored substance was quantitatively and parallel analyzed at a mass level, after 2200 Unigenes-containing cDNA array was hybridized with the <sup>33</sup>P-labelled 1<sup>st</sup>-strand cDNA probes RT-synthesized from the total RNAs at panicle booting and grain filling of rice (Oryza sativa L. Subsp. indica cv. Zhongzao 21). Expressed genes in filling were classified according to the ratio of normalized AR volumes of these two developmental stages, nearly half the efficient genes (694, 47.5%) were in house-keeping, that is no strong tissue-specific expressed (0.5&lt;R&lt;2) and one fourth of genes (350, 24.0%) were specific-highly expressed (R≥3) and involved in the accumulation of starch, protein and fat with the temporal-spatially expressed characteristics. Such related genes as sucrose synthase, starch synthase, Wx protein, branching enzyme, debranching enzyme, glutamine synthetase, proteinase inhibitor gene, polyubiquitin (RUBQ2) gene, 3-ketoacyl carrier protein synthase iii and stearyl-ACP desaturase, maybe took an important role in grain quality, taste, nutrition and flavor formation. Some genes related to seed tolerance and germination ability, for example, formic acid dehydrogenase, inhibitor-like protein, germin-like protein、GA regulated protein and α-amylase, also were monitored simultaneously.
