| Summary: | [Objectives] To investigate the effect of Astragalus polysaccharides (APS) combined with Ginsenoside Rg3 on PD-1/PD-L1 in colon cancer. [Methods] Fifty Balb/c mice were randomly divided into blank control group, model group, APS group, APS combined with Ginsenoside Rg3 group and 5-FU group, with 10 mice in each group. In addition to the blank group, subcutaneous transplantation tumor model of colon cancer was established in the other 4 groups. The tumorbearing mice in each group were subjected to continuous drug intervention for 12 days, during which the tumor volume and body weight of mice were measured and recorded daily. After the intervention, peripheral blood of mice was collected by removing eyeballs, and then the mice were killed by CO2 asphyxia. Spleen and tumor of mice were extracted and weighed. Real-time fluorescent quantitative PCR was used to detect the expression of PD-1/PD-L1 mRNA in tumor tissues of each group, immunohistochemistry was used to detect the expression of PD-1/PD-L1 in tumor tissues of each group, and ELISA was used to detect the expression of IL-6, TNF-α and IFN-γ in serum of mice in each group. [Results] The tumor volume in APS+Rg3 group and 5-FU group was lower than that in model group (P<0.05). There was no significant difference in tumor volume between APS group and model group (P>0.05). The spleen index of APS+Rg3 group was higher than that of model group, APS group and 5-FU group (P<0.001). There was no significant difference in spleen index between APS group and model group and 5-FU group (P>0.05). The mRNA expression levels of PD-1 and PD-L1 in APS+Rg3 group and 5-FU group were lower than those in model group and APS group (P<0.001). The mRNA expression levels of PD-1 and PD-L1 in APS+Rg3 group were not significantly different from those in 5-FU group (P>0.05). PD-1 is mainly expressed in cell membrane and is stained brown. PD-L1 is mainly expressed in the cell membrane or cytoplasm, and the stain is light yellow or brownish yellow. The expression level of PD-1 protein in APS group, APS+Rg3 group and 5-FU group was lower than that in model group (P<0.05). There was no significant difference in PD-1 protein expression between APS+Rg3 group and 5-FU group (P>0.05). The expression level of PD-L1 protein in APS+Rg3 group and 5-FU group was lower than that in model group (P<0.05). Compared with APS group, the expression level of PD-L1 protein in APS+Rg3 group and 5-FU group was decreased (P<0.05). The expression levels of IL-6, TNF-α and IFN-γ in APS+Rg3 group and 5-FU group were lower than those in model group and APS group (P<0.05). The expression level of IFN-γ in APS group was lower than that in model group, and the difference was statistically significant (P<0.05). There was no significant difference in the expression levels of IL-6 and TNF-α between APS group and model group (P>0.05). [Conclusion] APS combined with Ginsenoside Rg3 can inhibit the growth of colon cancer and PD-L1, improve the immune function of colon cancer mice, and reduce the expression levels of IL-6, TNF-α and IFN-γ. |
