| Summary: | In order to screen the high-yield β-caryophyllene (BCP) strains, the aroma components in light-flavor (Qingxiangxing) Baijiu and Daqu was detected by headspace solid-phase microextraction (HS-SPME)-GC-MS. The β-caryophyllene-producing strains were preliminarily screened by traditional isolation techniques, Fuqu was inoculated with primary screening strains to re-screening by β-caryophyllene content determination, and identified by molecular biology technique. The extraction conditions of β-caryophyllene in Fuqu were optimized, and the screened strains were applied in small tank fermentation. The results showed that the β-caryophyllene content in the light-flavor original liquor was 0.02 g/L, and the relative content of β-caryophyllene in Daqu was 0.85%. Five strains of mold (SDA1-SDA5) were preliminarily screened, and the strain SDA3 with the highest relative content of β-caryophyllene (1.04%) was identified as Rhizopus delemar. The β-caryophyllene content was 37.98 mg/L when the extraction reagent was anhydrous ethanol and the extraction time was 2 h. Compared with the addition of Daqu alone, the β-caryophyllene content in liquor produced by adding strain SDA3 to enhance Fuqu for small tank fermentation was increased from 13.65 mg/L to 24.65 mg/L. |
