| Summary: | Silver carp serum immunoglobulin was purified using immunoaffinity chromatographic column, and the serum IgM with high purity was then obtained. The IgM H chain and L chain were estimated to be about 76.4 kD and 27.2 kD, respectively. Simultaneously, mannan-binding protein (MBP) affinity chromatography, saturated ammonium sulfate (SAS) sedimentation and TG affinity chromatography methods were also used to purify the serum IgM. Although serum IgM was also successfully isolated by these methods, the purity attained differed, however, from that attained when the previous method was employed. It is considered that immunoaffinity method is more desirable in purification of fish serum immunoglobulin. |
